Supplementary Effects of Lentinus edodes with Different Harvest Period and Part on Neurotransmitters and Lipid peroxide levels in the Brain of Diabetic Mice

Interpretive Summary: In this paper, ARS scientist collaborated with scientists at the Rural Development administration of South Korean government to investigate the supplementary effects of Lentinus edodes in the brain of diabetic mouse model (KK mouse). The results showed that acetylcholine content was significantly higher in mice whose diet was supplemented with Lentinus edodes compared to untreated group. The activities of acetylcholinesterase and monoamine oxidase B enzyme were significantly inhibited in the brain of and lipid peroxide content was lower in mice group fed Lentinus edodes compared to control group. Furthermore, the cap of Lentinus edodes was most effectve in enhancing cognitive function compared to the other part of Lentinus edodes. These studies demonstarte important role of dietary supplementation with Lentinus edodes on brain function and has implication on the use of Lentinus edodes as a way to improve the brain health of people.

Technical Abstract: This study was designed to investigate the supplementary effects of Lentinus edodes which were harvested at different time period and part on acetylcholine content and its related enzyme activities in the brain of diabetic mouse model (KK mouse). We fed mice with standard diet(Control diet; CON) and 4 different kinds of experimental diets (DGC: on time harvested, cap of Dong Go; DGS: on time harvested, stipe of Dong Go; HSC: late harvested, cap of Hyang Sin; HSS: late harvested, stipe of Hyang Sin) to KK mouse for 8 weeks. Neurotransmitters such as acetylcholine contents, acetylcholinesterase activities, monoamine oxidase B activities and lipid peroxide contents in the brain were measured. The esults showed that acetylcholine content was significantly higher in DGC and HSC groups than CON group. The activities of acetylcholinesterase and monoamine oxidase B enzyme were significantly inhibited in the brain of DGC and HSC groups compared with CON group. Lipid peroxide content was lower in DGC group than CON group. These results suggested that the cap of Lentinus edodes which were harvested on time and late time contain increased acetylcholine content and decreased acetylcholinesterase activities, monoamine oxidase B activities and lipid peroxide contents. Thus the cap of Lentinus edodes which were harvested at different time periods may play an effective role in enhancing cognitive function.
